What PNC-27 Is

PNC-27 binds HDM-2 (the human homolog of MDM2) on the surface membrane of cancer cells — where HDM-2 is anomalously expressed compared to non-transformed cells. The peptide–HDM-2 complex inserts into the plasma membrane and forms transmembrane pores, leading to rapid colloid-osmotic necrosis of the cancer cell. Healthy cells do not display membrane HDM-2 and are reportedly spared. This is a necrosis-via-membrane-disruption mechanism, distinct from the apoptosis-induction mechanism of most p53-pathway therapeutics.

Michl, Bowne, and colleagues have published a series of pre-clinical papers reporting PNC-27 selectivity for cancer-cell lines (pancreatic, breast, leukemia, glioma) over non-transformed cells in vitro, and tumour-growth suppression in mouse xenograft models. No independent group has produced a major replication of the membrane-HDM-2 selectivity claim, and no human clinical trial has been registered.
